摘要: 油果樟(Syndiclis chinensis C. K. Allen)作为珍稀濒危物种,其叶绿体基因组研究对生物多样性保护具有重要意义。本工作通过高通量测序首次获得其完整叶绿体基因组(160 286 bp,GC含量39%),呈现典型环状四分体结构:LSC(89 335 bp)、SSC(24 623 bp)及IRs(各23 165 bp)。共注释出127个基因,含82个蛋白编码基因、37个tRNA和8个rRNA基因,其中44个基因参与光合作用,70个涉及自我复制,其余功能基因及未知基因各6个。SSR分析鉴定出235个多态性位点,单核苷酸重复占比73.6%(A/T主导)。系统发育树显示油果樟与同属物种形成高支持率进化分支,并与琼楠属亲缘关系较近。本研究首次揭示油果樟叶绿体基因组特征,为其种群遗传演化研究提供分子生物学依据。

Characterization and Phylogenetic Analysis of the Complete Chloroplast Genome of Syndiclis Chinensis

Abstract: Syndiclis chinensis C. K. Allen, a rare and endangered species, holds significant importance for biodiversity conservation. This study reports the first complete chloroplast genome of S. chinensis (160 286 bp, 39% GC content) obtained through high-throughput sequencing. The genome exhibits a typical circular quadripartite structure, comprising a large single-copy (LSC, 89 335 bp), a small single-copy (SSC, 24 623 bp), and two inverted repeat regions (IRs, 23 165 bp each). A total of 127 genes were annotated, including 82 protein-coding genes, 37 tRNA genes, and 8 rRNA genes. Functional classification revealed 44 photosynthesis-related genes, 70 self-replication genes, and 12 others (6 with other functions and 6 with unknown functions). Simple sequence repeat (SSR) analysis identified 235 polymorphic loci, with mononucleotide repeats (predominantly A/T) accounting for 73.6%. Phylogenetic analysis demonstrated that S. chinensis forms a highly supported evolutionary clade with congeneric species and is closely related to the Beilschmiedia genus. This study provides the first molecular insights into the chloroplast genome of S. chinensis, offering a foundation for elucidating its population genetics and evolutionary mechanisms, which is critical for conservation strategies.
