抵抗素、瘦素及脂联素在非酒精性脂肪肝大鼠中的变化

摘要: 探讨脂肪因子抵抗素、瘦素、脂联素在非酒精性脂肪肝(NAFLD)模型中的变化。将大鼠分为模型组和对照组,分别喂养高脂高胆固醇饲料和普通饲料4周。测定血清甘油三酯(TG)、胆固醇(TC)、谷丙转氨酶(ALT)、谷草转氨酶(AST),结合肝脏病理切片确定NAFLD模型的建立;测定大鼠血清中脂肪因子的水平,分析其与TGTCALTAST的相关性。模型组与对照组相比,血清中TGTCALTAST水平明显增加(P<0.05~0.01),肝小叶结构紊乱,肝细胞发生脂肪变性。模型组血清抵抗素和瘦素水平明显升高(P<001),脂联素水平明显降低(P<001)。血清瘦素与TGTC呈正相关,抵抗素与TGTCASTALT呈正相关,脂联素与TGTCASTALT呈负相关。抵抗素、瘦素和脂联素可能与NAFLD的发生发展有关。

Abstract: The purpose of this study is to investigate the changes of adipokines includig resistin, leptin and adiponectin in nonalcoholic fatty liver disease (NAFLD). The Rats were divided into model group and control group, and were fed high fat high cholesterol diet and normal diet for 4 weeks, respectively. Determination of serum triglyceride (TG), cholesterol (TC), alanine aminotransferase (ALT), aspartate aminotransferase (AST), combined with liver pathology to determine the establishment of NAFLD model. The levels of adipokines in serum of rats were measured, and the correlation between adipokines and TG, TC, ALT and AST was analyzed. The study found that compared with the control group, the levels of TG, TC, ALT and AST in the serum of the model group were significantly enhanced (P<005~001), the structure of the hepatic lobule was disordered, and the hepatic steatosis was observed. Serum resistin and leptin levels in the model group increased significantly (P<001) while adiponectin levels decreased significantly (P<001). serum leptin was positively correlated with TG and TC, resistin was positively correlated with TG, TC, AST and ALT, while adiponectin was negatively correlated with TG, TC, AST and ALT. Resistin, leptin and adiponectin may be correlated with the occurrence and progression of NAFLD.
