摘要: 采用响应面法优化乙醇回流提取山楂核总酚酸的最佳工艺条件并研究其降脂活性。实验考察乙醇浓度、回流时间、液料比、提取温度与提取次数5个因素对山楂核总酚酸提取率的影响。在单因素试验的基础上,采用响应曲面法中的BoxBehnken模式,对山楂核总酚酸的提取工艺条件进一步优化,并研究山楂核提取物的胰脂肪酶抑制活性。结果发现,山楂核总酚酸的最佳提取工艺为乙醇浓度70%,回流时间25 h,液料比22 mL·g-1,提取温度86 ℃,提取次数2次,在此条件下山楂核总酚酸提取率为483 mg·g-1。山楂核提取物在体外有良好的胰脂肪酶抑制活性,具有剂量依赖性。响应面优化的提取工艺稳定可靠,可用于山楂核总酚酸的提取,获得的总酚酸提取物具有良好的降脂活性。

Optimization of Extraction Process of Total Phenolic Acid from Crataegus Pinnatifida
Seeds by Response Surface Methodology and Its Lipidlower Activity

Abstract: The optimum conditions for total phenolic acid extraction from Crataegus pinnatifida seeds by ethanol reflux were optimized by response surface method and its lipidlowering activity was also studied. The effects of ethanol concentration, reflux time, liquidtoliquid ratio, extraction temperature and extraction times on the extraction rate of total phenolic acid from Crataegus pinnatifida seeds were investigated. Based on the single factor experiment, the BoxBehnken model in the response surface method was used to further optimize the extraction conditions of the total phenolic acid in the Crataegus pinnatifida seeds,and the inhibition on the pancreatic lipase activity of the Crataegus pinnatifida seeds extract was studied.The optimal extraction condition of total phenolic acid extraction from Crataegus pinnatifida seeds was ethanol concentration 70%, reflux time 25 h, liquidtoliquid ratio 22 mL·g-1, extraction temperature 86 ℃, extraction times 2 times.Under this condition, the total phenolic acid extraction rate of Crataegus pinnatifida seeds was 483 mg·g-1. Crataegus pinnatifida seeds extract has good pancreatic lipase inhibitory activity in a dosedependent manner. The response surface optimized extraction process is stable and reliable, and can be used for extraction of total phenolic acid from Crataegus pinnatifida nucleus.And the Crataegus pinnatifida seeds extract has good lipidlowering activity.
