摘要: 建立了大孔树脂纯化刺玫叶总黄酮的最佳工艺条件。以吸附率、解吸率为主要考察指标,采用静态吸附及解吸、动态吸附及解吸实验,确定了LX36型大孔树脂纯化刺玫叶总黄酮的效果较理想,并对其纯化条件进行了优化。其最佳纯化条件:上样量为m(生药)∶m(树脂)=1∶25,吸附速率为2 BV·h-1,静置15 min,吸附后用2 BV蒸馏水洗脱,再用5 BV的70%乙醇以3 BV·h-1的流速洗脱,在此最佳条件下,其吸附率平均可为9424%,解吸率平均为8859%,纯化后刺玫叶干浸膏中总黄酮含量由原来的821%提高到2512%,树脂富集倍数约为3倍。结果表明,采用LX36型大孔树脂对刺玫叶总黄酮的纯化具有一定效果。

Purification Technology of Total Flavonoids in Leaves of Rosa Davurica Pall.with Macroporous Resin Method

Abstract: The purification technology of total flavonoids(TF) in leaves of Rosa davurica Pall. with macroporous resin was optimized. Applying both static and dynamic absorptiondesorption experiments, the optimum purification conditions with LX36 macroporous resin which offered ideal purification function were determined using absorption ratio and desorption ratio of TS as evaluating indicators. The loading quantity of sample m(crude drugs)∶m(resin), adsorption rate, adsorption time, volume of water, ethanol content, desorption rate, the volume of eluant were 1∶25, 2 BV·h-1, 15 min,2 BV,70%, 3 BV·h-1, and 5 BV respectively, the average absorption and desorption rate were 9424% and 8859%. The purity of TF in extracts could be changed from 821% to 2512%, which offered about 3 times enrichment with this type of resins. The experimental results have shown that LX36 macroporous resin was fit for the purification of TS in leaves of Rosa davurica Pall.
