摘要: 在单因素实验基础上,以总黄酮提取率为考察指标,根据星点试验设计原理,利用响应面法建立数学模型,得到葫芦巴总黄酮提取的最佳工艺;采用DPPH自由基清除法评价葫芦巴总黄酮的体外抗氧化活性。葫芦巴总黄酮最佳提取工艺:乙醇体积分数68%,提取时间23 h,提取温度742 ℃,在此工艺条件下葫芦巴总黄酮提取率的预测值为214%,实际提取率为210%,与理论值间偏差为187%。葫芦巴总黄酮对DPPH自由基清除率随总黄酮浓度增加而上升,但对其清除能力弱于同浓度Vc。由响应面法得到响应面的葫芦巴总黄酮最佳提取工艺稳定,得到的葫芦巴总黄酮具有一定的体外抗氧化活性。

Extraction Process of Total Flavonoids from Trigonella Foenumgraecum and Its Antioxidant Activity in Vitro

Abstract: The optimum extraction conditions of total flavonoids from Trigonella foenumgraecum(TFF) and its antioxidant activities in vitro were investigated. Central composite design (CCD) based on the single factor experiment was used to design the experiment scheme. Response surface methodology (RSM) was used to analysis the experiment result and establish the mathematical model of extraction process of TFF with the extraction rate of total flavones as index. The antioxidant activity of TFF in vitro were evaluated by the methods of DPPH free radical scavenging. The optimum process was as follows: the concentration of ethanol 68%, extraction time 23 h, extraction temperature 742 ℃. Under the optimal condition, the predicted value of extraction rate was 214%, while the experimental value was 210%. The experimental value agreed with the predicted from the regression model with a relative error of 187%. Experimental results show that DPPH radical scavenging activity of TFF were enhanced with increasing concentration of TFF, but the scavenging capacity of TFF was weak compared with the same concentration of Vc. The optimum extraction process obtained by RSM is stable and TFF has some extent antioxidant activity in vitro.
