超临界流体-高效液相色谱法萃取芹菜中芹菜素

摘要: 芹菜素是芹菜中重要的活性化合物,具有抗癌、抗氧化、降血压的药用价值。传统的提取方法耗能高效率低,而超临界CO2流体萃取法可以有效的将芹菜素与其结构类似物分离,近年来在中草药活性成分的提取领域有重要应用。本研究使用CO2配以95%99%浓度的乙醇为辅溶剂对芹菜中的芹菜素进行萃取,并通过高效液相色谱(HPLC)对其含量进行测定,以探究超临界萃取芹菜素的最佳条件。结果表明:添加辅溶剂乙醇可以显著提高芹菜素的萃取率,当辅溶剂乙醇浓度为95%时芹菜素的萃取率最高,可达0006%。在整个萃取过程中,前120 min萃取效率最高,约有90%的芹菜素被萃取出来,当萃取时间持续到150 min时,芹菜素已基本萃取完全。

Abstract: Apigenin is an important active compound in celery which has significant medicinal value. In this study, the supercritical fluid extraction method was used to extract apigenin from celery using carbon dioxide and ethanol with 95% and 99% concentration as the auxiliary solvent, and its content was determined by high performance liquid chromatography (HPLC) to explore the optimal conditions for supercritical extraction of apigenin. The results showed that the extraction rate of apigenin could be significantly increased by adding cosolvent ethanol, and the extraction rate of apigenin was the highest when the concentration of cosolvent ethanol was 95%, up to 0006%. In the whole extraction process, the extraction efficiency was the highest in the first 120 min, about 90% apigenin was extracted. When the extraction time lasted to 150 min, apigenin was basically extracted completely.
