延髓背侧迷走复合体到下丘脑弓状核ghrelin神经通路的构成

摘要: 探讨延髓背侧迷走复合体(DVC)到下丘脑弓状核(ARCghrelin神经通路的构成并观察ghrelin受体(GHSR1a)在ARC的表达。采用逆行追踪结合荧光免疫组化染色的方法,观察DVCARCghrelin免疫阳性神经元的纤维投射;采用聚合酶链反应(Polymerase Chain ReactionPCR)结合荧光免疫组化染色技术,观察GHSR1aARC的定性及定位表达。DVC发出ghrelin阳性神经元的纤维投射至ARC,并呈现出同侧投射的优势。ARCGHSRla mRNA和蛋白的表达。DVCARC存在ghrelin神经通路,该通路的功能有待进一步研究。

Composition of the Ghrelin Neural Pathway from the Dorsal Vagal Complex to the Arcuate Nucleus of the Hypothalamus

Abstract: To investigate the composition of the ghrelin neural pathway from the Dorsal vagal complex (DVC) to the Arcuate nucleus (ARC) and observe the expression of growth hormone secretagogue receptor1a (GHSR1a) in ARC. Retrograde tracing andimmunofluorescence histochemical staining were used to observe the fiber projection of ghrelin immunopositive neuronfrom DVC to ARC. Polymerase chain reaction (PCR) andimmunofluorescence histochemical staining were used to observe the qualitative and localized expression of GHSR1a in ARC. DVC emitted fibers of ghrelinpositive neurons which projected to ARC and exhibited the advantage of ipsilateral projection. ARC had GHSRla mRNA and protein expression. There is a ghrelin neural pathway from DVC to ARC, and the function of this pathway needs further study.
