摘要: 使用MTT比色分析法、台盼蓝染料排除测定法、流式细胞分析技术和蛋白免疫印迹法,探讨了PIP对肺癌细胞的抗肿瘤活性及分子机制。研究结果显示,用PIP对人肺癌细胞株(A549和NCIH23)进行处理,显著抑制细胞增殖并诱导凋亡。研究还发现PIP显著诱导A549和NCIH23细胞凋亡,并与Bcl2、Bclxl和survivin的蛋白水平降低及活化型caspase3的水平升高有关。同时,PIP也可抑制磷脂酰肌醇3激酶(PI3K)的表达以及Akt和mTOR信号蛋白的磷酸化。研究结果表明:PIP不仅可以抑制肿瘤细胞增殖,而且可通过调节PI3K/Akt/mTOR信号通路诱导肿瘤细胞凋亡,提示荜茇酰胺可能是一种潜在的抗肺癌天然产物。

Piperlongumine Induces Apoptosis of Lung Cancer Cells
via the PI3K/Akt/mTOR Signaling Pathways

Abstract: In the present study, using MTT colorimetric analysis, trypan blue dye exclusion assay, flow cytometry and Western blotting to report the antitumor effects of PIP on lung cancer cells and the action of potential mechanism. Treatment of human lung cancer cells (A549 and NCIH23) with PIP inhibited the proliferation and induced apoptosis in both cell lines as characterized by the increased Annexin Vbinding. The induction of apoptosis by PIP was associated with a decrease in the levels of Bcl2, Bclxl, survivin, and activated caspase3. Further,treatment with PIP also inhibited the expression of phosphatidylinositol3kinase (PI3K), phosphorylation of Akt and mTOR signaling proteins. Our study demonstrates that PIP not only inhibits cell growth but also induces apoptosis through modulation of the PI3K/Akt/mTOR pathway in human lung cancer cells. It suggests that PIP may be a potentail candidate agent against lung cancer.
