超临界萃取山楂核总黄酮的研究

摘要: 研究超临界萃取法提取和大孔树脂纯化山楂核总黄酮的工艺条件。以山楂核为原料,采用超临界CO2萃取技术萃取山楂核总黄酮,采用紫外分光光度法对萃取物中的总黄酮进行定量分析,用正交设计实验法考察了萃取温度、时间、压力和夹带剂用量对提取总黄酮的影响;并用大孔树脂对总黄酮提取物进行纯化,考察了乙醇浓度、体积和流速对大孔树脂纯化总黄酮的影响。结果发现,超临界萃取的最佳工艺条件为:液固比为10 mL·g-1、萃取温度50 ℃、压力20 MPa和时间2 h,总黄酮得率可达2827%。大孔树脂纯化总黄酮的最佳工艺条件:70%乙醇、体积为60 mL、流速15 mL·min-1,总黄酮得率可达532%。该提取方法和纯化条件简单易操作,可达到废物利用的目的。

Abstract: Supercritical extraction of total flavonoids from hawthorn seeds and purification by macroporous resin were studied. Hawthorn seeds were tested as raw materials, total flavonoids of hawthorn seeds were extracted by supercritical CO2 extraction technique. The total flavonoids in the extract were quantitatively analyzed by the ultraviolet spectroscopy method, and the effects of extraction temperature, time, pressure and the dosage of the entrainer on total flavonoids yield were observed by orthogonal design experiment method. The total flavonoids were purifed by macroporous resin and the effects of ethanol concentration, ethanol volume and ethanol flow rate on the total flavonoids have been investigated. The experimental results showed that the best technological conditions for supercritical extraction were liquidsolid ratio 10 mL·g-1, extraction temperature 50 , extraction pressure 20 MPa, and extraction time 2 h. Under this condition, the yield of total flavonoids was 2827%. The best technological conditions of purification in macroporous resin were the 70% ethanol, the volume of 60 mL and the flow rate of 15 mL·min-1, and the yield of total flavonoids was 532%. This extraction and purifying method is easy to operate, and can realize the goal of cyclic utilization.
