摘要:采用加热回流提取、超声提取、微波提取以及加压溶剂提取等4种方法提取马齿苋黄酮,比较不同方法所得马齿苋黄酮提取率和DPPH·清除率的差别,并采用扫描电镜观察提取后药材微观结构的改变,在此基础上利用正交设计法对加压溶剂提取工艺进行优化。结果表明:加压溶剂法提取所得马齿苋黄酮提取率最高。其DPPH·清除率比另3种提取方法略高;最优提取工艺为以70%乙醇为溶剂,压力0.20 MPa,时间30 min,料液比1∶45 (g·mL-1),温度80℃,此条件下提取率为1.33%,该工艺重现性好,可为工业上提取马齿苋黄酮提供参考。

Comparison and Optimization of Techniques for Extraction of Flavonoids from the Portulacae Herba

Abstract: Flavonoids from Portulacae herba were extracted by reflux extraction, ultrasonic assisted extraction, microwave assisted extraction, and pressurized solvent extraction(PSE). The extraction yield of flavonoids and DPPH scavenging capacity of the extracts from 4 methods were compared. Scanning electron microscope images of the plant cells after four methods treatment were obtained to provide visual evidence of the extraction effect. The results show that the extraction yield of flavonoids by PSE was the highest, and the extracts obtained with PSE also showed the best antioxidant activity. The optimum conditions of PSE were 70% ethanol as solvent, temperature 80℃, solid-liquid ratio 1∶45 (g·mL-1), extraction time 30 min, pressure 0.20 MPa, and on this condition the flavonoids yield was 1.33%. It was suggested that PSE could be used to extract the flavonoids from portulacae herba for the food or pharmaceutical industries.
