摘要: 采用乙醇回流法提取柚皮中总黄酮,研究其最佳工艺条件及其抗氧化活性。在单因素实验基础上,选择液料比、乙醇浓度、回流时间3因素,采用星点设计响应面法优化提取工艺条件。采用邻苯三酚法测定柚皮提取液的体外抗氧化活性。星点实验结合生产实际,乙醇回流提取的最佳工艺条件为液料比20∶1(mL∶g),乙醇浓度76%,回流时间95 min,回流温度90 ℃,提取2次。该条件下总黄酮得率预测值为1032%,实测平均值为1036%,RSD为072%。柚皮提取液具有良好的抗氧化活性,并呈现出一定的剂量依赖关系。

Optimization of Extraction Conditions of Total Flavonoids from Pomelo Peel Using Response Surface Methodology and Its Antioxidant Activity

Abstract: Total flavonoids from pomelo peel were extracted by ethanol reflux extraction, and the optimal conditions for extraction and its antioxidant activity were determined in this experiment. Three extraction parameters including liquidtosolid ratio, ethanol concentration and reflux time were identified as main factors affecting the extraction yield of total flavonoids. The extraction parameters were optimized by a central composite designresponse surface methodology (CCDRSM). Antioxidant activity of pomelo peel extract was measured by Pyrogallol method in vitro. The results combined with practice revealed that the optimal conditions for total flavonoids extraction from pomelo peel were 95 min extraction twice with 76% ethanol at a liquidtosolid ratio of 20∶1(mL∶g) and reflux temperature of 90 ℃. Under these conditions, the predicted and experimental extraction yields of total flavonoids were 1032% and 1036% respectively and RSD was 072%. The antioxidant activity in vitro experiments showed that pomelo peel extract had good antioxidant activity, and exhibited a certain dosedependent relationship.
