基于转录调控网络研究的乳腺癌与系统性红斑狼疮免疫系统发病机理探寻

 摘要:利用快速独立成分分析(FastICA)方法提取乳腺癌(BC)和系统性红斑狼疮(SLE)2种疾病的差异表达基因。在此基础上,结合转录因子对靶基因调控的生物学先验信息,利用网络成分分析(NCA)算法提取了转录因子在2种疾病中调控强度及对靶基因调控活性的变化情况,构建2种疾病均有显著变化的11个转录因子和18个靶基因的转录调控网络。该研究为进一步探寻BC和SLE的免疫发病机制和相应的分子生物学实验提供了新的方向和依据。

 Exploration of Pathogenesis in Immune System of Breast Cancer and Systemic Lupus Erythematosus Based on Transcriptional Regulation Network Study

 Abstract: Independent component analysis (FastICA)was applied to extracting the differentially expressed genes between breast cancer (BC) and systemic lupus erythematosus (SLE). On this basis, this article extracts the regulation intensity of transcription factors(TFs)in two diseases and the changes of activity of transcription factors regulating target genes through network component analysis (NCA) algorithm and construct the transcriptional regulatory network of 11 transcription factors and 18 target genes which has significant changes in two diseases combined with the biology priori knowledge that transcription factors regulate target genes. The study provides a new direction and basis for further exploration of immune pathogenesis of BC and SLE and corresponding molecular biology experiments.
