摘要: 采用热水浸提法提取丹参叶中总酚酸,优化提取工艺并检测提取物对酪氨酸酶的抑制作用。以总酚酸得率为指标,采用星点设计响应面法考察液料比、提取温度和提取时间对提取工艺的影响,采用多元线性回归和二项式方程拟合,确定最佳提取工艺。以左旋多吧为底物,观察丹参叶总酚酸提取物对酪氨酸酶的抑制作用。实验结果表明:丹参叶总酚酸的最佳工艺条件为液料比23∶1,提取时间109 min,提取温度76 ℃。该条件下总酚酸得率预测值为3.211%,实测平均值为3.285%。丹参叶总酚酸提取物可抑制酪氨酸酶的活性,并具有一定的剂量依赖关系。星点设计响应面法可用于丹参叶总酚酸的提取工艺优化,预测性良好;获得的提取物具有一定的美白效果。

Optimization of Extraction of Total Phenolic Acids from Leaves of Radix Salvia
Miltiorrhiza Using Response Surface Methodology and Its Inhibition on Tyrosinase

Abstract: Total phenolic acids from leaves of Radix salvia Miltiorrhiza were extracted by hot water extraction, and the optimal conditions for extraction were determined and its inhibition on tyrosinase was observed. The effects of liquidtosolid ratio, extraction temperature and extraction time on the total phenolic acids yield were observed using central composite designresponse surface methodology (CCDRSM). The extraction condition was optimized by multiple linear regression and binomial equation. The inhibition on tyrosinase of total phenolic acids extracts was measured by the utilization of LDA as substrate. The results revealed that the optimal condition for total phenolic acids from leaves of Radix salvia Miltiorrhiza were as follows: Liquidtosolid ratio 23∶1, extraction time 109 min and extraction temperature 76 ℃. Under these conditions, the predicted and experimental extraction yields of total phenolic acids were 3211% and 3285% respectively. The total phenolic acids extracts inhibited the tyrosinase activity with a certain dosedependent manner. CCDRSM could be used for optimization of extraction technology of total phenolic acids from leaves of Radix salvia Miltiorrhiza with good predictability and the extracts obtained shows a whitening effect.
