新型吡嗪甲酰-L-苯丙氨酰-L-亮氨腈/缬氨腈二肽的合成

摘要: 利用混合酸酐法和催化氢化经6步反应,合成了4种新型吡嗪甲酰-L-苯丙氨酰-L-亮氨腈/缬氨腈二肽,对反应条件进行了优化,利用IR、1H NMR、13C NMR和HR-MS对其结构进行了表征。结果表明:对于混合酸酐法,反应温度0℃,THF为溶剂,三乙胺为缚酸剂,n(氨基酸)∶n(Et3N) ∶n(ClCOOEt) = 1∶1.5∶1.5时,缩合反应的产率最高可达90%;对于催化氢化反应,以甲醇为溶剂,10%Pd/C为催化剂,催化剂的质量分数为30%,反应时间为30 min时,氢化反应产率最高可达90%。该法原料价廉易得,操作简便,反应条件温和,产率较高。

Synthesis of Novel Dipeptide of Pyrazinylformyl-L-phenylalanyl-L-leunitrile/Valnitrile

Abstract:  Four novel dipeptides of pyrazinylformyl-L-phenylalanyl-L-leunitrile/valnitrile compounds were synthesized by mixed-carbonic anhydride method and catalytical hydrogenation through a six-step reaction.The structures of the obtained compounds were characterized by IR, 1H NMR, 13C NMR and HR-MS. The synthetic procedures have been optimized for mixed-carbonic anhydride method and cataytical hydrogenation. The yield of the product of mixed-carbonic anhydride method achieved 90% under the  optimal condition of temperature 0 ℃, THF as solvent, Et3N as acid binding agent, n (amino acid)∶n(Et3N)∶n(ClCOOEt)=1∶1.5∶1.5. And the yield of catalytical hedrogenation achieved 90% while methanol as solvent,10%Pd/C as catalyst, 30% (mass fraction) catalyst was added and reacted for 30 min by the results. The method has advantages, such as the ready availability of the starting materials, easy workup procedure, mild reaction conditions, and moderate to good yields.
