壳聚糖药用纳米粒子的制备及性能研究

 摘要: 应用壳聚糖与甲基丙烯酸甲酯(MMA)通过自由基聚合反应,以硝酸铈铵为引发剂合成一种功能新颖的药用纳米粒子,作为药物的载体材料,与药物结合后可以调节药物释放速度,减少给药次数,从而增加药物治疗的安全性、高效性和可靠性。研究结果表明,硝酸铈铵可以较好地引发接枝反应,当反应温度为50 ℃,反应时间为4 h,壳聚糖0.6 g、甲基丙烯酸甲酯6 mL、硝酸铈铵0.9 g、1%醋酸300 mL时,合成的纳米粒粒径最小,平均粒径在179 nm左右。体外溶出实验表明,当n(阿司匹林)∶n(壳聚糖)=1∶1时,制备出的纳米粒载药量、包封率较高,可达72.1%,载药纳米粒在16 h内对药物有良好的缓释作用,具有较好的应用前景。

 Preparation and Properties of Medical Chitosan Nanoparticles

 Abstract: Chitosan and methyl methacrylate(MMA) were prepared into novel-functional medical nanoparticles through radical polymerization. Nanoparticles loaded with medicine can regulate degree of releasing, and reduce deliver time, in order to increase safety, high-effect and dependment of medication as a carrier material of medicine. It is showed that ammonium ceric nitrate  is a good initiator in graft reaction. The grain diameter of compounded nanoparticle is minimized into 179 nm on average when temperature is 50 ℃, reacting time 4 h, chitosan 0.6 g, MMA 6 mL, ammonium ceric nitrate 0.9 g, and 1% acetic acid 300 mL. The vitrorelease test shows that the drug-loading rate and envelop rate is up to 72.1%, and nanoparticles works well in sustained-releasing medicine when the proportion of aspirin and chitosan reaches 1∶1.
