基于适体和连续复制的生物传感器对凝血酶的检测

摘要:  利用DNA适体对靶物质的识别,以及DNA聚合反应具有替换模板链上结合的靶目标的特点,设计了一个分子机器用于适体靶的检测。适体序列通过酰胺键连接CdS纳米颗粒,当溶液中出现待检测的凝血酶的时候,使引物互补部分暴露,在DNA聚合酶、dNTP(4种三磷酸脱氧核苷酸混合物)组成的聚合体系的作用下,引发一系列聚合反应,将大量附有凝血酶和CdS纳米颗粒的适体序列替换下来释放到溶液中。溶液中的CdS纳米颗粒可以通过阳极溶出伏安法(ASV)测定。本研究通过这一方法探讨作为靶的凝血酶的存在和浓度与ASV响应的关系,从而建立这一检测方案的定量关系,并探讨这一检测方案对于其他类似物质的选择性。

 Detection of Thrombin with a Biosensor Based on Aptamers and Continuous Replication

 Abstract: In this study, CdS nanoparticles were prepared as electrochemical markers, and were labeled with the 5′end amino-modified aptamer DNA sequences using the condensation reaction to produce the CdS nanoparticle-labeled DNA probes with the electrochemical activity. The probes were immobilized on magnetic beads through DNA hybridization. In the polymerization system, a specific recognition of the thrombin aptamer will trigger polymerization, which replaces the tag CdS probe and releases it into the solution. In the polymerization and replacement, one thrombin molecule was repetitively employed as target, amplifying the signal to a great extent. The Anodic Stripping Voltammetric (ASV) interrogation of CdS different nucleotides were tested using this system to show the good selectivity of this system.
