摘要: 探讨了小茴香挥发油纳米粒的制备工艺及其体外抗肿瘤活性。以尤特奇S100为载体材料,采用纳米沉淀法制备小茴香挥发油纳米粒并用单因素实验法优化制备工艺。用透射电镜、马尔文粒径仪观察纳米粒形态并测定其粒径和电位;用透析法测定其体外释放;考察不同温度下纳米粒的稳定性;用MTT实验、划痕实验和Transwell实验分别检测小茴香挥发油纳米粒对人结肠癌RKO细胞增殖、迁移和侵袭的影响。结果发现,小茴香挥发油纳米粒的最优制备工艺条件为:V(有机相)∶V(水相)为1∶2、ES100浓度为14 mg·mL-1、搅拌速度为1 500 r·min-1、V(小茴香挥发油)∶V(ES100)比为4∶7。制备的小茴香挥发油纳米粒形态良好,包封率和载药量高,在pH 74环境中释放,可于结肠局部发挥作用;25 ℃环境下放置5周稳定性良好。同时还发现,小茴香挥发油纳米粒能显著抑制结肠癌RKO细胞的增殖、迁移和侵袭。

Preparation and Antitumor Activity of Nanoparticles
Loaded with Fennel Volatile Oil

Abstract: The preparation and antitumor activity of nanoparticles loaded with Fennel volatile oil were investigated in this study. Using Eudragit S100 (ES100) as the carrier material, nanoparticles loaded with Fennel volatile oil were prepared by nano-precipitation method and the preparation technology was optimized by single factor experiment. The morphology of nanoparticles was observed by transmission electron microscope and Malvern partilce size meter. The release in vitro was measured by dialysis method. The stability of nanoparticles at different temperatures was investigated. The effect of nanoparticles loade with Fennel volatile oil on proliferation, migration and invasion of human colon cancer RKO cells were detected by MTT assay, scratch assay and Transwell assay, respectively. The results showed that the optimal preparation conditions were as follows: The volume ratio of organic phase to aqueous phase was 1∶2, the concentration of ES100 was 14 mg·mL-1, the stirring speed was 1 500 r·min-1, and the dosage ration of Fennel volatile oil to ES100 was 4∶7. The prepared nanoparticles loaded with Fennel volatile oil had good morphology, high encapsulation rate and drug loading, and can be released in the enviroment of pH74 that was suitable to play a role in the colon. The nanoparticles were stable for 5 weeks at 25 ℃. It was also found that nanoparticles loaded with Fennel volatile oil could significantly inhibit the proliferation, migration and invasion of colon cancer RKO cells. This study provides an important reference for the preparation development and pharmacological application of fennel volatile oil.
