中华绒螯蟹组织蛋白酶B基因的SNPs及其与鳗弧菌抗性/易感性的关系

摘要: 组织蛋白酶B作为一种溶酶体半胱氨酸样蛋白酶,在中华绒螯蟹的抗病原体感染中起着重要的作用。本研究克隆了EsCatB基因序列,并对其多态性进行了鉴定。从60个个体中鉴定出19个单核苷酸多态性(SNP)位点,均为同义突变。通过对抗性组和易感组中等位基因频率和基因型频率的分布差异进行统计分析,表明EsCatB编码区1SNP(852 C/T)与中华绒螯蟹对鳗弧菌的抗性显著相关(P<0.01)SNP(852 C/T)位点的基因型频率(P=0.003)和等位基因频率(P=0)在易感组和抗性组间差异显著。具有TT基因型和T等位基因的中华绒螯蟹对鳗弧菌具有较高的抗性。因此,EsCatB编码序列中的SNP位点(852 C/T)可作为一种分子标记,促进中华绒螯蟹抗鳗弧菌的选择选育。

with Resistance/Susceptibility to Vibrio Anguillarum

Abstract: As a lysosomal cysteine like-protease, cathepsin B of the Chinese mitten crab Eriocheir sinensis (EsCatB) plays crucial roles in defense against pathogen infection. In this study, the sequence of EsCatB was cloned and its polymorphisms were identified. Nineteen single nucleotide polymorphism (SNP) loci were identified from 60 individuals (30 susceptible and 30 resistant), and they were all synonymous mutations. Statistical analysis of the distribution difference of allele frequency and genotype frequency between resistant and susceptible groups indicated that one SNP (852 C/T) in the coding region of EsCatB was significantly associated with the resistance of E. sinensis against Vibrio anguillarum (P<0.01). The SNP 852 C/T genotype frequency (P=0.003) and allele frequency (P=0) differed significantly between the susceptible and resistant groups. E. sinensis with the TT genotype and T allele were highly resistant to V. anguillarum. Thus, the SNP locus (852 C/T) in the coding sequence of EsCatB potentially could be used as a molecular marker to facilitate the selective breeding of E. sinensis resistant to V. anguillarum.
