摘要: 利用ATP适体修饰磁珠为分离载体,以电化学试剂结晶紫(CV)适体、CV、ATP适体互补序列修饰的胶体金CV/DNA2/DNA3/AuNPs为探针,建立了测定ATP电化学新方法。方法检测限为10×10-9 mol·L-1,另外,该方法对ATP的测定表现出了良好的选择性。并利用该方法对运动前后小鼠心肌ATP含量进行了测定。

Determination of ATP in Cardiac Musculus Based Magnetic Bead Separation and DNA Labeled Gold Nanoparticles as Probe

Abstract: ATP aptamer was immobilized onto the surface of magnetic beads. Then the probe which was labeled with aptamer complementary sequence and crystal violet aptamer was hybridizated onto it. The introduction of ATP induced release of the probe. The electrochemical signal was produced from the released probe. The experimental results demonstrated it can be to detect 10×10-9 mol·L-1 ATP. The method also showed excellent selectivity toward GTP,UTP CTP. The protocol was successfully applied to the determination of ATP in cardiac musculus.
