摘要: 采用蔗糖溶液在-120 ℃下将聚异戊二烯聚苯乙烯共聚物(PIPS)的薄膜材料进行包埋,然后用超薄切片机制成了50 nm厚的超薄切片,并用透射电子显微镜清晰地观察到了所制薄膜中纳米尺度的两相分离结构。与用环氧树脂和聚酯等树脂包埋聚合物薄膜的方法相比,用蔗糖溶液低温包埋聚合物薄膜的方法具有简便、快速、安全、高效、成本低和环保等诸多优点。

One Simple Physical Embedding Technique of Polymer Film for Cryoultramicrotomy

Abstract: We provided a new embedding method of polymer film for cryoultramicrotoming. Polyisoprene(PI)polystyrene(PS) copolymer film was embedded with frozen sucrose solution at-120 ℃and then microtomed into ultrathin sections with a thickness of 50 nm. It was found that the phase separation morphology of thus prepared ultrathin film can be observed clearly by transmission electron microscope(TEM). In comparison with the embedding methods of polymer film with resins like epoxy and polyester, the sucrose solution embeding method displayed such advantages as convenience, speediness, safety, efficiency, low cost, environment protection and so on.
