PLA-PEG对斑马鱼补体免疫系统的影响

 摘要: 通过实时定量PCR测定补体系统中主要补体因子的表达情况,探究了PLA-PEG(聚乳酸-聚乙二醇共聚物)在生物体内对免疫系统的影响。向成年斑马鱼腹腔注射PLA-PEG溶液,同时向对照组注射灭菌水,在注射后1,3,5,7,9,11 d后分别取肝脏,提取RNA,反转录得到cDNA,使用RT-PCR仪器测定补体(C1r/s、C3、C4、C6、Bf、MASP、MBL)的表达量,并分析其在注射后的变化量。结果表明,注射后各补体因子表达量有不同程度的上调,说明PLA-PEG可以激活生物体内补体系统。

  The Influence of PLA-PEG on Zebrafish Complement System

 Abstract: We discussed the immune process. of PLA-PEG(poly lactic acid polyethylene glycol copolymer)in vivo by determining the expression levels of complement factor by Real-time PCR. Method: Use the mature zebrafish as exprimental material, and intraperitoneal injection of PLA-PEG, while the control group is injected with sterile water. Take the liver of zebrafish at 1,3,5,7,9,11d post injection, then extract RNA and reverse transcriptase into cDNA.To determinate the expression levels of complement (C1r/s, C3, C4,C6, Bf, MBL,MASP) using Real-time PCR, we analyzed the change of expression levels after injection. The result showed all the complement factor expressing different levels increased after injection, and this could be considered the PLA-PEG can activate the immune response of complement system.
