摘要: 从土壤样品中筛选出高产抗生素(Zwittermicin A)的苏云金芽孢杆菌菌株:A164,对此菌种的发酵条件进行了探讨。以A164合成高含量抗生素为目标对发酵培养的碳源、氮源、时间等条件进行了单因素优化;采用Plackett-Burman设计,确定发酵条件的重要影响因子,根据爬坡路径法,确定了各个因子的最适范围,采用响应面分析法对因子再次进行优化,得到了最佳发酵条件。最佳发酵条件为:蛋白胨含量17.56 g·L-1,葡萄糖20.0 g·L-1, MgSO4 0.18 g·L-1,CaCl2 0.08 g·L-1,ZnSO4 0.20 g·L-1,K2HPO4 1.3 g·L-1,培养温度28.31 ℃,发酵时间36 h。

Optimization of Fermentation Conditions of Bacillus Thuringiensis
for High Production of Zwittermicin A

Abstract: Bacillus thuringiensis A164 for high production of Zwittermicin A was separated from the soil samples,then the fermentation conditions of this strain was discussed. According to single factorial experiments, carbon source, nitrogen source, and time were optimized. Plackett-Burman design was used to evaluate the importance of the related influential factor. Then the path of steepest ascent experiment was adopted to realize the optimal region of these importance factors. At last, the optimal fermentation conditions were obtained by response surface methodology as followed: 17.56 g·L-1 of soybean powder, 20.0 g·L-1 of glucose, 0.18 g·L-1 of MgSO4, 0.08 g·L-1 of CaCl2, 0.20 g·L-1 of ZnSO4 , 1.30 g·L-1 of K2HPO4 , temperature 28.31 ℃, and fermentation time 36 h.
