脂肪酶在复合模板剂下氨基改性介孔SiO2中的固定化活性

 摘要:  以三嵌段聚合物(P123)和表面活性剂聚乙二醇(PEG 4000)为复合模板剂,使用水热法合成了高度有序的介孔SiO2 (MS),并用3-氨丙基三乙氧基硅烷(APTES)对其进行后接枝改性。通过XRD、SEM、TEM和氮气吸脱附等手段对介孔SiO2进行了表征。结果表明,该介孔材料具有高度有序的六方状介孔结构。当介孔材料经表面氨基改性后,其孔径、孔体积和比表面积均相应降低,但其形貌和介孔结构仍保持不变。以该材料为载体对假丝酵母脂肪酶(CRL)固定化,考察了改性前后酶固定量和固定化酶的催化活性。结果显示改性后介孔SiO2固定化酶具有较好的催化活性和重复利用性,重复利用5次后,该固定化酶仍具有其最初活性的50%以上。

 Immobilization of Lipase on Amino-functionalized Mesoporous Silica Prepared Using a Composite Template

 Abstract: Mesoporous silica (MS) was synthesized via hydrothermal process using triblock copolymer Pluronic P123 and poly (ethylene glycol) (PEG 4000) as dual templates. The materials were functionalized with (3-aminopropyl)triethoxysilane APTES by post-synthesis-grafting method. The results indicated that mesoporous silica possessed a 2-dimensional hexagonal mesoscopic structure. The mesoscopic structure could be remained, while pore diameters, pore volumes, and surface areas were decreasing after the functionalization. Candida rugosa lipase (CRL) was used as a model enzyme for studying the effect of amino-functionalized in loading amount and enzymatic activity. As a result, APTES functionalized mesoporous silica immobilized CRL showed higher hydrothermal stability and the greatest reusability, and its residual activity was maintained at levels of 50% to the initial value after the fifth reuses.
