摘 要:采用开环聚合后修饰的方法合成了一种聚氨基酸,它能够在水中自组装成聚合物胶 束,由于这种聚合物胶束对酯酶具有响应性,使得它能够实现对肿瘤细胞的选择性细胞毒作 用。在酯酶响应方面,加入酯酶后可以维持胶束的棒状形态实现药物的原位缓慢释放,在细 胞实验中,负载了 DOX 的胶束在药物浓度为 6. 3 μg · mL−1 时,癌细胞 Hela 细胞存活率仅为 10% 左右,正常细胞 L929 细胞的存活率则在 80% 以上。实验结果证明,合成的载药胶束可以 与肿瘤细胞中丰富的酯酶响应,有效地杀死肿瘤细胞而对正常细胞的杀伤力很小,提高了化 疗药物的利用率并大大降低了化疗药物的副作用。

Preparation of Esterase-Responsive Drug-Loaded Micelles and Its Application in Chemotherapy

Abstract: A polyamino acid was synthesized by ring-opening polymerization and postmodification. It can self-assemble into polymer micelles in water. Because this polymer micelle is responsive to esterase, it can achieve selective cytotoxicity to tumor cells. In terms of ester⁃ ase response, the addition of esterase can maintain the rod-like morphology of micelles to achieve in-situ slow release of drugs. In cell experiments, when the drug concentration of DOX-loaded micelles was 6. 3 μg · mL−1 , the survival rate of cancer cells Hela cells was only about 10%, and the survival rate of normal cells L929 cells was above 80%. The experimental results show that the synthesized drug-loaded micelles can respond to the rich esterase in tumor cells, effectively kill tumor cells and have little lethality to normal cells, improve the utilization of chemotherapy drugs and greatly reduce the side effects of chemotherapy drugs.
