纳米酶增强的喷涂式水凝胶用于细菌感染的糖尿病创面皮肤再生

摘要: 高血糖和细菌感染引起的慢性炎症以及氧化应激微环境严重阻碍了糖尿病患者的创面皮肤再生。本研究制备了一种具有自修复和喷涂性能的多功能水凝胶(PAM)用于调控糖尿病创面微环境以期达到皮肤再生的目的。PAM水凝胶网络由氨基酸聚合物(poly (K-co-C))和银离子(Ag+)通过配位作用形成,Ag+的存在实现了对革兰氏阳性菌和革兰氏阴性菌的双重杀伤。另外,将二氧化锰纳米颗粒(Alb-MnO2)掺入PAM水凝胶以用于消耗创面处的内源性过氧化氢(H2O2),重塑氧化还原微环境。实验结果表明,PAM 水凝胶在细菌感染的糖尿病小鼠模型中很好地促进了创面皮肤再生。

Abstract: Chronic inflammation caused by hyperglycemia and bacterial infection as well as the oxidative stress microenvironment severely hinder the regeneration of traumatized skin in diabetic patients. This study prepared a multifunctional hydrogel (PAM) with self-healing and sprayable properties for regulating the microenvironment of diabetic wounds with the aim of promoting skin regeneration. The PAM hydrogel network is formed by the coordination of amino acid polymers (poly (K-co-C)) and silver ions (Ag+), and the presence of Ag+ achieves dual killing of Gram-positive and Gram-negative bacteria. In addition, we incorporated manganese dioxide nanoparticles (Alb-MnO2 into PAM hydrogels to deplete endogenous hydrogen peroxide (H2O2 at the trauma site and reshape the redox microenvironment. The experimental results showed that the PAM hydrogel promoted the regeneration of traumatic skin well in the diabetic mouse model of bacterial infection.
