摘 要:小鼠灌胃给予不同剂量的葡萄柚精油(分别为 0. 1、0. 2、0. 4 mg ·(10 g)−1 ),腹腔注射 四氯化碳(CCl4)诱导急性肝损伤,于实验结束时取血清和肝脏,制备肝匀浆和肝组织石蜡切 片。使用试剂盒测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、肿瘤坏死因子-α(TNF-α)、 白介素-6(IL-6)和肝组织超氧化物歧化酶(SOD)、丙二醛(MDA)的含量,肝脏石蜡切片行苏 木素-伊红染色。采用 Western blot测定肝组织中核转录因子-κB(NF-κB)的表达。结果发现, 葡萄柚精油显著减轻 CCl4引起的肝损伤,降低肝指数以及血清中 ALT、AST、TNF-α、IL-6 以 及肝组织中的 MDA 水平,升高肝组织中的 SOD 水平,抑制肝组织 NF-κB 的表达。葡萄柚精 油可能通过抗炎抗氧化活性发挥对急性肝损伤小鼠的保护作用。

Protective Effect of Grapefruit Essential Oil on Acute Liver Injury in Mice

Abstract:The aim of this study was to investigate the protective effect of grapefruit essential oil on acute liver injury in mice and explore the possible mechanism. Mice were given different doses of grapefruit essential oil (0. 1,0. 2,0. 4 mg ·(10 g)−1 body weight, respectively) by intragastric administration. Intraperitoneal injection of carbon tetrachloride (CCl4) was con⁃ ducted to establish acute liver injury model. Serum and liver were taken at the end of the experiment. Liver homogenate and paraffin section of liver tissue were prepared. Serum alanineaminotransferase (ALT), Aspartate aminotransferase (AST), tumor necrosis factor (TNF- α), interleukin-6(IL-6), and hepatic superoxide dismutase (SOD) and malondialdehyde (MDA) were determined by kits. Hematoxylin-eosin staining (HE) was performed in paraffin sections of liver, and the expression of nuclear transcription factor-κB (NF-κB) in the liver was deter⁃ mined by Western blot. The results showed that grapefruit essential oil significantly alleviated the liver injury induced by CCl4, reduced liver index, serum ALT, AST, TNF- α, IL-6 and hepatic MDA, while increased hepatic SOD, and inhibited the expression of NF- κB in the liver. Grapefruit essential oil has a protective effect on acute liver injury in mice via antiinflammatory and antioxidant activities.
